The effects of 2,4-diaminopyridine (2,4-DAP) on high K(+)-induced contraction and histamine release were investigated in isolated guinea-pig jejunal segments. Concentration-response curves to histamine were constructed in the presence and absence of 2,4-DAP. In Ca(2+)-free medium, high K+ (25 mM) induced contraction in all segments, and this contraction was abolished by 2,4-DAP (10-100 microM). In the presence of 1 mM Ca2+, the high K(+)-induced contraction was greatly potentiated, and this potentiation was abolished by 2,4-DAP (10-100 microM). In the presence of Ca2+, histamine concentration-response curves were constructed in segments from the guinea-pig jejunum in the presence and absence of 2,4-DAP (1-100 microM). At a histamine concentration of 1 microM, contraction was induced in all segments. In addition, a further increase in the histamine concentration induced contraction, which was accompanied by a parallel shift to the right in the concentration-response curve. However, the contraction induced by a high concentration of histamine was abolished by 2,4-DAP (10 microM). These findings suggest that high K(+)-induced contraction and histamine release are attenuated by 2,4-DAP and the attenuating effects are inhibited by the stimulation of an H2-receptor.
